I am about to add a refug to my main tank. Once the refug is set up will this cause a spike in my main tank.

Unless your planning on using LS that has been sitting a bucket of nasty water for a month, or adding uncured LR it should do absolutely nothing negative. I'm assuming your going to be adding sand, maybe some LR, and macro algae? If you do add rock, just make sure it's cured first.

^^ I agree...You'll be fine depending on what you add to the fuge. Let us know what exactly you plan to add to it

As stated above, as long as you're using live sand and live rock that's already cycled, you'll have no issues. The refugium will also give you the added benefit of being able to fill it up with Chaeto, which is great a nitrate absorber.
